A practical reference on semax: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.
Reviewed 2026-08-01. Anything still debated is marked as such rather than presented as settled.
Semax binds to melanocortin receptors and is thought to influence neuronal survival and plasticity rather than to act through the adrenal axis. Laboratory work has shown increased expression of brain-derived neurotrophic factor and nerve growth factor in treated tissue. Changes in c-Fos, a marker of neuronal activation, have also been reported. Because the peptide is rapidly degraded by peptidases, its effects are generally attributed to downstream signaling cascades rather than to sustained receptor occupancy.
Published studies are dominated by animal models of stroke, ischemia, and cognitive impairment, with a smaller number of human trials conducted in Russia. Many of the human reports are small, single-center, and published in Russian-language journals, which limits independent scrutiny. Outcome measures vary between studies and often rely on clinician-rated scales rather than objective biomarkers. Systematic reviews have noted the methodological weaknesses and called for larger, preregistered trials before firm conclusions can be drawn.
Claims about enhanced focus, memory, or mood in healthy people rest mostly on anecdotal reports and community discussion rather than on controlled data. It remains unclear whether any cognitive benefit observed in patients recovering from brain injury would extend to uninjured users. Dose-response relationships, long-term safety, and interactions with other drugs are not well characterized in the peer-reviewed literature. Questions about optimal route of administration and treatment duration likewise remain open.
Stability depends heavily on physical state. Lyophilized powder held dry, cold and dark retains its content over long periods, whereas dissolved peptide begins to change within days at room temperature. The most cited degradation route is oxidation of the methionine residue, which converts the peptide to a sulfoxide form that elutes differently on chromatography. Hydrolysis of amide bonds and adsorption onto container walls contribute smaller losses. Buffers that exclude oxygen from the headspace slow the oxidation pathway, but no single condition prevents all change indefinitely.
Practical handling follows from those properties. Bulk material is best divided into single-use portions soon after receipt, because each thaw exposes the whole container to moisture and temperature cycling. Vials should be allowed to reach room temperature before opening to prevent condensation on the powder. Low-binding plasticware reduces loss of dilute solutions, and sterile filtration is used when a preparation must remain free of microbial growth. Records of batch number, reconstitution date and storage history are what allow a later analytical result to be interpreted meaningfully.

Proposed mechanisms center on neurotrophic signaling rather than on classical melanocortin receptor activation. Rodent experiments have reported shifts in the expression of brain-derived neurotrophic factor and nerve growth factor after administration, together with changes in the associated receptor systems. Several authors argue that the peptide acts largely through its degradation products and their interaction with peptidergic pathways, but this remains a hypothesis rather than a settled finding. No single molecular target has been identified in a way that the field broadly accepts.
Published research covers ischemic stroke, traumatic brain injury, cognitive impairment, optic nerve conditions and attention-related measures. Much of the human evidence comes from small trials conducted in one country, which limits how far the results generalize. Animal models supply the larger share of the data, and effects seen in rodents do not transfer automatically to people. Reviews have noted that methodological reporting is often incomplete, making it difficult to pool results or compare treatment schedules across studies.
Pharmacokinetic accounts emphasize rapid breakdown. After intravenous dosing the intact peptide disappears from blood within minutes, and nasal delivery produces low but measurable concentrations. Metabolites rather than the parent molecule may account for part of the observed activity, although the relative contribution is unresolved. Dosing in the literature varies widely and no optimal schedule has been agreed. These gaps are regularly cited as a reason the findings have not produced broad clinical adoption beyond the original research setting.
The mechanisms attributed to semax are inferred from animal and cell studies rather than traced to one confirmed target. The most frequently cited pathway involves increased expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal and cortical tissue. Some work points to engagement of melanocortin receptors, particularly MC4, which the parent ACTH fragment can activate. Effects on dopaminergic and serotonergic signalling have also been reported. No single account explains all observed results, and the relative weight of each pathway remains unsettled.
Published studies examine a fairly narrow set of endpoints. Rodent experiments commonly measure maze learning, infarct volume after induced ischemia, and tissue levels of neurotrophic factors. Clinical reports from Russian centres describe attention, memory and recovery scores in patients after stroke or transient ischemic attack. Most of those human studies are small and few have been repeated by independent groups. Outcome measures differ between studies, which limits direct comparison.
Circulation time for the peptide is short because peptidases cleave it readily. The Pro-Gly-Pro tail is thought to slow breakdown compared with the bare ACTH fragment, but the gain appears modest. Absorption after intranasal dosing is limited, and only a fraction of a dose is expected to reach the central nervous system. Laboratory concentrations therefore sit well above levels achieved systemically, a gap that complicates translation from bench findings to clinical claims.
There are several medications classes that are commonly used to control blood sugar levels in patients with diabetes. Most of the medications used are either oral or injected. In patients with T1D, insulin is required because the body no longer produces insulin. In patients with T2D, management is largely more variable as lifestyle changes can have a significant impact. However, medications may be added to further help control blood glucose levels if the lifestyle changes are not effectively controlling the condition. Unlike patients with type 1 diabetes, patients with T2D can still produce insulin, so usually oral medications are recommended as first line treatment before requiring insulin for diabetic control. Patient education and adherence with treatment is very important in managing the disease. Improper use of medications and insulin can be very dangerous causing hypo- or hyper-glycemic episodes.
=== Ivory Lady === In 2008 the skeleton of a high-ranking individual buried in the site's tomb was discovered and was thought to be a man. Because of surrounding ivory artifacts, the skeleton has been dubbed the Ivory Man. However, in 2021 it was announced that, based on the analysis of sex-specific amelogenin peptide in tooth enamel, the skeleton is that of a woman. The corresponding results were published in 2023 by Nature and a new name for the skeleton, the Ivory Lady, was proposed.
After participating in various roles in the company, Snyder assumed the presidency in 2010, becoming the company's sixth president. However, most major decisions are made by a seven-member executive team. Snyder does not intend to franchise nor sell and plans to pass on ownership of the company to her children. In 2018, In-N-Out donated $25,000 to the California Republican Party. In 2021, it donated $40,000. In-N-Out COO Mark Taylor and his wife, Traci, have donated to Donald Trump's presidential campaigns. On January 24, 2024, In-N-Out announced the closure of its only store in Oakland, California, due to safety concerns related to crime, marking the first time in its 75-year history that the company has shut down a location. On August 1, 2026, a mass shooting occurred at In-N-Out's Twin Falls, Idaho restaurant, killing three and wounding seven. The gunman, 24-year old Chad Williams, was confronted by Jordan Salinas, an armed civilian, and an off-duty state trooper. Williams eventually committed suicide. The motives for the attack are still unknown. The restaurant had opened about a week earlier.
== Discovery == Notch was discovered in a mutant Drosophila in March 1913 in the lab of Thomas Hunt Morgan. This mutant emerged after several generations of crossing out and back-crossing beaded winged flies with wild type flies and was first characterized by John S. Dexter. The most frequently observed phenotype in Notch mutant flies is the appearance of a concave serration at the most distal end of the wings, for which the gene is named, accompanied by the absence of marginal bristles. This mutant was found to be a sex-linked dominant on the X chromosome that could only be observed in heterozygous females as it was lethal in males and homozygous females. The first Notch allele was established in 1917 by Charles William Metz and Calvin Bridges. In the late 1930s, studies of fly embryogenesis done by Donald F. Poulson provided the first indication of Notch's role in development. Notch mutant males exhibited a lack of the inner germ layers, the endoderm and mesoderm, that resulted in failure to undergo later morphogenesis embryonic lethality. Later studies in early Drosophila neurogenesis provided some of the first indications of Notch's roll in cell-cell signaling, as the nervous system in Notch mutants was developed by sacrificing hypodermal cells. Starting in the 1980s researchers began to gain further insights into Notch function through genetic and molecular experiments.

A lobotomy (from Greek λοβός (lobos) 'lobe' and τομή (tomē) 'cut, slice') or leucotomy is a discredited form of neurosurgical treatment for psychiatric disorder or neurological disorder (e.g. epilepsy, depression) that involves severing connections in the brain's prefrontal cortex. The surgery severs most of the connections to and from the prefrontal cortex, and the anterior part of the frontal lobes of the brain. From the 1930s until the 1970s, the treatment was used for handling psychiatric disorders as a mainstream procedure in some countries. A preoccupation with the ability to work and personal responsibility over patient well-being were contributing factors to the prevalence of lobotomies in the United States. The originator of the procedure, Portuguese neurologist António Egas Moniz, shared the Nobel Prize for Physiology or Medicine of 1949 for the "discovery of the therapeutic value of leucotomy in certain psychoses", although the awarding of the prize has been subject to controversy. The procedure was modified and championed by Walter Freeman, who performed the first lobotomy at a mental hospital in the United States in 1936. Its use increased dramatically from the early 1940s and into the 1950s; by 1951, almost 20,000 lobotomies had been performed in the US and proportionally more in the United Kingdom. More lobotomies were performed on women than on men: a 1951 study found that nearly 60% of American lobotomy patients were women, and limited data shows that 74% of lobotomies in Ontario from 1948 to 1952 were performed on female patients.
==== Superficial vein thrombosis ==== SVT is the development of a thrombus in a superficial vein. SVT is not normally clinically significant, but the thrombus can migrate into the deep venous system where it can also give rise to a pulmonary embolism. The main risk factor for SVT in the lower limbs is varicose veins.
=== Varieties and cultivars === Lotus varieties have been classified according to their use into three types: rhizome lotus, seed lotus, and flower lotus. Varieties that show more than one of these characteristics are classified by the strongest feature. Regarding production area in China, rhizome lotus has the largest area with 200,000 ha (490,000 acres), followed by seed lotus with 20,000 ha (49,000 acres).

=== Menopausal hormone therapy === CPA is used at low doses in menopausal hormone therapy in combination with an estrogen to provide endometrial protection and treat menopausal symptoms. It is used in menopausal hormone therapy under the brand name Climen, which is a sequential preparation that contains 2 mg estradiol valerate and 1 mg CPA. Climen was the first product for use in menopausal hormone therapy containing CPA to be marketed. It is available in more than 40 countries.
=== Tuberous sclerosis complex === Sirolimus also shows promise in treating tuberous sclerosis complex (TSC), a congenital disorder that predisposes those afflicted to benign tumor growth in the brain, heart, kidneys, skin, and other organs. After several studies conclusively linked mTOR inhibitors to remission in TSC tumors, specifically subependymal giant-cell astrocytomas in children and angiomyolipomas in adults, many US doctors began prescribing sirolimus (Wyeth's Rapamune) and everolimus (Novartis's RAD001) to TSC patients off-label. Numerous clinical trials using both rapamycin analogs, involving both children and adults with TSC, are underway in the United States.
Access to transition care, mental care, and other issues affecting transgender people is very limited; there is only one comprehensive transgender health care clinic available in South Africa. Additionally, the typical lack of access to transition options that comes as a result of gatekeeping is compounded by the relatively limited knowledge of transgender topics among psychiatrists and psychologists in South Africa.
== Career == Armstrong began his academic career at Rosalind Franklin University of Medicine and Science, where he served as Professor of Surgery and Associate Dean at the Scholl College of Podiatric Medicine. He later moved to Tucson and accepted an offer at the University of Arizona. In 2017, Armstrong was honored as University Distinguished Outreach Professor of Surgery—the first ever in the history of the University of Arizona's Department of Surgery. Following the University of Arizona, Armstrong accepted a position at the Keck School of Medicine of the University of Southern California's Department of Surgery, where he was later named Distinguished Professor of Surgery and Neurological Surgery. In 2025, USC bestowed upon Armstrong the title of Distinguished Professor—one of the university's highest academic honors—making him the first Doctor of Podiatric Medicine to receive this distinction at USC.

It is associated with the melanocortin receptor family, particularly subtypes found in the central nervous system. This interaction is separate from the adrenal pathway activated by full-length ACTH.
Peptidases degrade the compound within minutes in circulation. This short half-life is one reason nasal delivery is used, placing the substance close to the olfactory and trigeminal pathways.
No large multicenter trials have been published in English-language journals. The existing human evidence consists mainly of smaller studies conducted in Russia, which limits generalization.
Suppliers normally quote a percentage of total chromatographic peak area, most often from reversed-phase HPLC. That figure says nothing about what the remaining percentage contains, and it depends on the detection wavelength used. A mass spectrometry result is a separate and stronger check on identity.
