A study compared different particle sizes of oral micronized estradiol. A preparation with the smallest particles (mainly <0.6 μm) was found to have the most rapid absorption and the highest bioavailability. However, a sharp peak in estradiol levels, without an accompanying rise in estrone levels, was observed during the first 2 hours with this particle size. It was suggested that the smallest estradiol particles may have been absorbed by the lymphatic system, partially bypassing first-pass metabolism and resulting in very high initial estradiol levels. The preparations with the larger particle sizes (mainly <3.5 μm and <20 μm) were found to be absorbed more slowly, without a pronounced initial peak in estradiol levels. Levels of estradiol were more even and similar to physiological levels with these particle sizes. Differences in area-under-the-curve estradiol levels with the different particle sizes were relatively small. As such, micronization may improve absorption but does not necessarily improve therapeutic effect. Micronized estradiol is rapidly and completely absorbed with oral administration. This is true for oral doses of 2 mg and 4 mg, but absorption was found to be incomplete for an oral dose of 8 mg. This dose showed 76% of the expected bioavailability based on dose proportionality and area-under-the-curve levels, indicating a small deviation from linearity. The absolute bioavailability of oral micronized estradiol is approximately 5%, with a possible range of 0.1% to 12%. As such, the bioavailability of oral estradiol is very low even with micronization.
== Secretion == A typical adult human stomach will secrete about 1.5 liters of gastric juice daily. Gastric juice is the combination of gastric gland secretions including the main component of hydrochloric acid (gastric acid), gastric lipase and pepsinogen. Once in the stomach pepsinogen is changed by gastric acid to the digestive enzyme pepsin adding this enzyme to the gastric juice. In humans, the pH of gastric acid is between one and three, much lower than most other animals, but is very similar to that of carrion eating carnivores, needing extra protection from ingesting pathogens. Gastric acid secretion is produced in several steps. Chloride and hydrogen ions are secreted separately from the cytoplasm of parietal cells and mixed in the canaliculi. This creates a negative potential of between −40 and −70 mV across the parietal cell membrane that causes potassium ions and a small number of sodium ions to diffuse from the cytoplasm into the parietal cell canaliculi. Gastric acid is then secreted along with other gland secretions into the gastric pit for release into the stomach lumen. The enzyme carbonic anhydrase catalyses the reaction between carbon dioxide and water to form carbonic acid. This acid immediately dissociates into hydrogen and bicarbonate ions. The hydrogen ions leave the cell through H+/K+ ATPase antiporter pumps. At the same time, sodium ions are actively reabsorbed. This means that the majority of secreted K+ (potassium) and Na+ (sodium) ions return to the cytoplasm.
=== European settlement === The first European settlement in South Africa was founded at the Cape of Good Hope in 1652, and administered as part of the Dutch Cape Colony. As a result of political turmoil in the Netherlands, the British occupied the Cape three times during the Napoleonic Wars, and the occupation became permanent after the Battle of Blaauwberg in 1806. The colony was then home to about 26,000 colonists settled under Dutch rule. Most represented old Dutch families brought to the Cape during the late 17th and early 18th centuries. Broadly speaking, the colonists included distinct subgroups, including the Boers. The Boers were itinerant farmers who lived on the colony's frontiers, seeking better pastures for their livestock. Many were dissatisfied with aspects of British administration, in particular with Britain's abolition of slavery in 1834. Boers who used forced labor were unable to collect compensation for their slaves. Between 1836 and 1852, many elected to migrate away from British rule in what became known as the Great Trek. Around 15,000 trekking Boers departed the Cape Colony and followed the eastern coast towards Natal. After Britain annexed Natal in 1843, they journeyed farther north into South Africa's eastern interior. There, they established two independent Boer republics: the South African Republic (1852; also known as the Transvaal Republic) and the Orange Free State (1854).

It has a formula of C29H20ClN7O11S3 and a molecular weight of 774.2 g/mol. Cibacron blue is soluble in water and DMSO, however insoluble in ethanol. In water, saturated concentration is reached at 12.92 mM with the help of sonication. Cibacron Blue F3GA has a wide specificity for nucleotide-binding proteins or just a stereoselectivity electrostatic binding. It can be used to purify interferons, dehydrogenases, kinases, and serum albumin. For example, interferon purification from human gingival fibroblast extract using Cibacron Blue F3G-A on poly(2-hydroxyethyl methacrylate), the supporting matrix, in the form of cryogels. It has shown 97.6% purity of interferon.

As of 2021, the drug epidemic in the United States was the deadliest it had ever been, according to federal data. More than 100,000 people died of drug overdoses in the United States during the 12-month period ending April 2021, according to provisional data published November 17, 2021, by the US Centers for Disease Control and Prevention. Overdose deaths increased 28.5% from the same period a year earlier and nearly doubled over the previous five years. Opioids continued to be the primary cause of drug overdose deaths. Additionally, the drug is increasingly affecting younger populations. A 2018 study found that fentanyl is involved in the majority of opioid-related deaths and that deaths involving fentanyl were more likely to occur in younger age groups and among non-Hispanic white individuals. Furthermore, young adults are increasingly affected by nonfatal fentanyl overdoses in recent time periods along with these other deadly occurrences. The drug is 50 to 100 times stronger than morphine and often cut with other drugs, meaning the user does not know they are taking fentanyl. The Drug Enforcement Administration (DEA) says 2.2 pounds represents half a million lethal doses. Synthetic opioids, primarily fentanyl, caused nearly two-thirds (64%) of all drug overdose deaths in the 12-month period ending April 2021, up 49% from the year before, the CDC's 's National Center for Health Statistics found. Fentanyl, a synthetic opioid painkiller, is 50 to 100 times more potent than morphine and 30 to 50 times more potent than heroin, with only 2 mg becoming a lethal dose.

The first arc of the series, Trails in the Sky, consists of three games. They are set within the Liberl Kingdom and primarily follow Estelle Bright and her adopted brother Joshua, members of the Bracer's Guild, a civilian peacekeeping and monster-hunting organization. The first game, Trails in the Sky, was released in Japan in 2004 for Windows and for the PlayStation Portable (PSP) in 2006. The second game, Trails in the Sky SC, continues the plot of the first and was released in Japan for Windows in 2006 and the PSP the following year. The third game, Trails in the Sky the 3rd, follows Septian Church agent Kevin Graham, who is sent to an otherworldly dimension known as Phantasma alongside many characters from the previous two games. Sky the 3rd also features vignettes exploring character backstories as well as foreshadowing plot elements explored in later arcs. It was released in Japan in 2007 for Windows and the PSP in 2008. A 3D remake of the first game, Trails in the Sky 1st Chapter, released worldwide for Nintendo Switch, Nintendo Switch 2, PlayStation 5, and Windows in 2025.
