A practical reference on ACTH(4-10): what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.

The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.
Reported pharmacological work centers on neurotrophic signaling, including changes in BDNF and NGF expression in hippocampal tissue in animal models. Human data come largely from studies conducted in Russia, and how well those results generalize to other populations remains an open question. Regulatory status differs sharply by jurisdiction: Semax is a registered prescription medicine in Russia, while it holds no approved marketing status in the United States or the European Union. Outside such jurisdictions it is generally handled as a research chemical, which affects both documentation and quality expectations.
Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. It was developed as a fragment analog of adrenocorticotropic hormone, modeled specifically on the ACTH(4-10) region. The first four residues reproduce that fragment, while a Pro-Gly-Pro tripeptide is appended at the C-terminus. Work on the compound originated in Russia, where it entered clinical use as an intranasal preparation. Its sequence places it among short regulatory peptides studied for effects on the central nervous system rather than on the adrenal axis.
Identity and purity are established with standard peptide methods. Reversed-phase HPLC with ultraviolet detection near 214 nm resolves the parent peak from deletion and truncation byproducts, and reports typically quote a main-peak percentage. Mass spectrometry by electrospray or MALDI-TOF confirms the expected molecular mass, while amino acid analysis or peptide mapping can verify composition when the sequence itself is in question. A certificate of analysis that pairs a chromatogram with a mass spectrum is more informative than a purity figure alone. Counter-ion content and residual solvents are separate specifications and are frequently omitted.
Semax is normally supplied as a lyophilized powder, and that form is the most stable. Suppliers commonly recommend storage at -20 °C in a desiccated container protected from light, with short-term handling at room temperature limited to weighing and transfer. Powder that has absorbed moisture degrades faster, so vials should be warmed to ambient temperature before opening to prevent condensation on the contents. Hygroscopic behavior is typical of short hydrophilic peptides, and humidity control matters more than temperature alone for long-term retention.
Once dissolved, the peptide is considerably less stable than the solid. Aqueous solutions are usually prepared at neutral to slightly acidic pH, filtered, and divided into single-use aliquots before freezing. Repeated freeze-thaw cycles are a common cause of avoidable loss and are best prevented by never refreezing a thawed aliquot. Adsorption to plastic and glass surfaces can lower the measured concentration of dilute solutions, particularly below roughly 0.1 mg/mL. Buffer choice, salt content, and container material all influence how much peptide remains detectable after storage.

The compound was developed during the 1980s at the Institute of Molecular Genetics in Moscow as part of research on fragments of adrenocorticotropic hormone. Early work examined short ACTH-derived sequences that retained neurotrophic effects while lacking the endocrine activity of the full hormone. Semax entered clinical use in Russia during the 1990s, where it received registration for several neurological indications. Outside that region it remained primarily a laboratory research material rather than an approved therapeutic. English-language literature on it grew more slowly and frequently cited the original Russian studies.
Terminology around the compound varies by source. It appears in catalogues and papers as Semax, as the heptapeptide ACTH(4-7)-Pro-Gly-Pro, and under various alphanumeric laboratory codes used by individual suppliers. These names refer to the same sequence but may imply different salt forms, purity grades, or counter-ions. Peptide databases usually list the free base mass, while product descriptions sometimes report acetate or trifluoroacetate salts with a different formula weight. Because naming conventions for research peptides are not standardised across vendors, checking the declared sequence and measured mass is more reliable than relying on a trade name alone.
Published research has focused mainly on neurological and cognitive endpoints in animal models, with proposed mechanisms involving brain-derived neurotrophic factor and related signalling pathways. A substantial share of the human data originates from a limited number of research groups, and independent replication in other countries remains sparse. Regulatory status reflects that distribution: the peptide is registered as a medicine in Russia and appears in some neighbouring markets, while elsewhere it is handled as a research chemical without approved therapeutic labelling. Questions about dose-response relationships, long-term effects, and comparability across studies are still open.
Lyophilised powder is normally kept at -20 °C in a desiccated container, with some suppliers recommending -80 °C for long-term archival storage. Repeated freeze-thaw cycles are the most common cause of avoidable loss, so aliquoting before freezing reduces variability between working sessions. Dissolved peptide is far less stable than the dry solid and is usually prepared fresh or held briefly at 4 °C. Aqueous solutions support both hydrolysis of the backbone and oxidation of the N-terminal methionine, and these two routes dominate degradation under ordinary laboratory conditions.
Pharmacological accounts link semax to melanocortin signalling and to modulation of neurotrophic factor expression, particularly brain-derived neurotrophic factor and nerve growth factor. Much of this evidence comes from rodent studies using intranasal delivery, a route chosen because it allows peptides to reach the central nervous system with limited systemic exposure. Whether the same mechanisms operate in humans at comparable magnitude remains an open question. The precise receptor or receptors responsible for the reported behavioural and neuroprotective effects have not been conclusively identified.
Clinical reports describe use in ischaemic stroke, transient ischaemic attack, optic nerve conditions, and cognitive complaints, but most of these studies are small and were conducted in a single region. Systematic reviews have generally described the evidence base as limited in size and variable in methodological quality. Randomised controlled data suitable for international regulatory assessment are scarce. As a result, major treatment guidelines outside Russia do not include the peptide, and interest in it remains largely research-driven rather than routine clinical.
=== Cleaning behavior === Atlantic blue tangs engage in cleaning behaviors with other fish as both cleaners and clients. In these interactions, cleaners remove parasites and other materials from the surface of the client. Clients benefit by having ectoparasites and damaged tissue removed from the outside of their body. Removal of unwanted organisms and tissue can lead to improved health maintenance.

Unlike aerated chocolates produced with large bubbles, micro-aerated chocolate has nitrogen beaten into the mixture. The resulting chocolate, different not only in bubble size but also flavour and texture. This is theorised to arise from the gas having a different solubility. Aerated chocolates containing long tubes of air are also created using this technique, but their distinction comes from how the aerated chocolate is extruded rather than set in a moulded shell. The final type, those held together by a skeleton of solid particles, are produced using water evaporation methods. In the first of these, sugar is dissolved in water. This solution is emulsified with cocoa liquor and milk solids, which is evaporated and then dried using industrial machinery, until the product contains less than 2% water. In the second, water is emulsified with liquid chocolate and set and then freeze dried, removing the water.
As the father of classical utilitarianism, Jeremy Bentham (1748–1832) proposed the felicific calculus as a method to measure, compare, and aggregate degrees of pleasure and pain, considering factors such as their intensity and duration. He also suggested a direct link between well-being and right action, arguing that individuals and governments should promote "the greatest happiness of the greatest number". His student John Stuart Mill (1806–1873) modified Bentham's idea, proposing that the contribution to well-being depends on the quality of the pleasure. He held that higher pleasures of the mind are more valuable than lower pleasures of the body, even if they have the same intensity and duration. Thomas Carlyle (1795–1881) rejected the utilitarian pursuit of happiness and the individualist market economy associated with Adam Smith, arguing instead that a sense of community and meaningful work are essential to well-being. The emergence of psychology as an empirical science prompted research into the internal factors of well-being, such as William James's (1842–1910) introspective research into the connection between happiness and religious experience. The psychoanalytic theory of Sigmund Freud (1856–1939) characterized the pursuit of pleasure and the reduction of tension as central aims of life. His early collaborator Carl Jung (1875–1961) explored how well-being depends on individuation as a form of psychological growth in which conscious and unconscious aspects are integrated.
John Lawrence, the Lord Mayor of London, ordered that the bodies of plague victims "...shall be at least six foot deep." The city officials apparently believed this would inhibit the spread of the disease, not realising that the true vector was fleas living on rats in the streets. In the event, there were so many victims that very few were buried in individual graves. Most were placed in massive plague pits so it is unlikely that this event alone gave rise to the "six feet" tradition.

Emopamil binding protein is a protein that in humans is encoded by the EBP gene, located on the X chromosome. EBP was discovered through its high-affinity binding to anti-ischemic drugs such as emopamil, from which it also derives its name. In addition to emopamil, EBP also bind with high affinity a variety of structurally unrelated compounds, such as amiodarone, opipramol, ifenprodil, trifluoperazine, and chlorpromazine. EBP has a mass of 27.3 kDa and resembles the σ2-receptor that resides in the endoplasmic reticulum of various tissues as an integral membrane protein.
Compton rule An empirical law of physical chemistry which states that the heat of fusion of a given element multiplied by its atomic weight and then divided by its melting point in kelvin is always equal to approximately 2.
=== March === 6 March Astronomers report the discovery of GJ 887 d, a super-Earth exoplanet orbiting the nearby red dwarf star GJ 887. Detected using radial velocity measurements, the planet lies within the star's habitable zone. At just 10.7 light-years, this becomes the second-nearest known exoplanet in the habitable zone after Proxima Centauri b. The UK's first long‑distance robotic-assisted surgery is reported to have been performed on a patient located 1,500 miles (2,400km) away in Gibraltar. 9 March – A study published in Geophysical Research Letters reports with statistical confidence that global warming has accelerated since around 2015, with the rate of increase rising from 0.2 °C to about 0.35 °C per decade after accounting for natural variability. The findings suggest the 1.5 °C threshold could be reached before 2030 if current trends continue. 10 March – A study by Vattenfall and Spoor using AI-assisted video monitoring of 2,007 bird flight paths near an offshore turbine at Aberdeen Bay over 19 months reports no confirmed collisions, with observed avoidance behaviour suggesting lower-than-expected impacts of wind turbines on seabirds. 11 March Two separate studies, published in Environmental Research Letters and Nature Communications, show how net negative emissions will need to be sustained for centuries in order to stabilise global climate change. Astronomers report evidence of a likely collision between two planets orbiting the star Gaia20ehk, about 11,000 light years away in the constellation Puppis.

Compartmental models are a mathematical framework used to simulate how populations move between different states or "compartments". While widely applied in various fields, they have become particularly fundamental to the mathematical modelling of infectious diseases. In these models, the population is divided into compartments labeled with shorthand notation – most commonly S, I, and R, representing Susceptible, Infectious, and Recovered individuals. The sequence of letters typically indicates the flow patterns between compartments; for example, an SEIS model represents progression from susceptible to exposed to infectious and then back to susceptible again. These models originated in the early 20th century through pioneering epidemiological work by several mathematicians. Key developments include Hamer's work in 1906, Ross's contributions in 1916, collaborative work by Ross and Hudson in 1917, the seminal Kermack and McKendrick model in 1927, and Kendall's work in 1956. The historically significant Reed–Frost model, though often overlooked, also substantially influenced modern epidemiological modeling approaches. Most implementations of compartmental models use ordinary differential equations (ODEs), providing deterministic results that are mathematically tractable. However, they can also be formulated within stochastic frameworks that incorporate randomness, offering more realistic representations of population dynamics at the cost of greater analytical complexity.
=== May === 1 May: 5,000 senior doctors affiliated with the Association of Salaried Medical Specialists strike in response to a pay dispute with national health service Health New Zealand. == Public debate and criticisms == The plan to build the integrated resorts was subject to considerable debate among Singaporeans even until 2014. Several groups, especially those belonging to religious communities as well as social workers, openly expressed their disapproval of the casinos. Concerns were raised about the negative social impact of casino gambling, citing worries that the casinos could encourage more gambling and increase the risk of compulsive gambling. Activist groups argued that a casino could also lead to undesirable activities often associated with gambling, including money laundering, loan sharks or even organized crime. Lee acknowledged the downsides of having integrated resorts and the concerns expressed by the public. He promised that there would be safeguards to limit the social impact of casino gambling. He stated there would be restrictions on the admission of local people into the casinos. Lee announced a steep entrance fee of S$100 per entry or S$2,000 per year (increased to $150 per entry or $3,000 per year on 4 April 2019) and a system of exclusions for all Singaporeans. In addition, the casinos would not be allowed to extend credit to the local population who are not premium players. Premium players are players who opened a deposit account with the casino and has a credit balance of at least SGD $100,000. The six-month consultative period gave the opportunity for many sections of the population to voice their opposition to the casinos, including a petition hat attracted tens of thousands of signatures.
== Thiol specificity == GPx-3 has a wide thiol specificity. The sources of reducing power for GPx-3 in vitro include GSH, cysteine, mercaptoethanol, and dithiothreitol. There is an evidence of effectiveness of homocysteine in reduction of GPx-3: GSH can be completely replaced by reduced homocysteine in vitro.

The sequence is Met-Glu-His-Phe-Pro-Gly-Pro, a seven-residue peptide. The first four residues correspond to the ACTH(4-10) fragment, and Pro-Gly-Pro is added at the C-terminus. N-acetylated versions also appear in the literature and differ in mass.
Semax is a synthetic analog of the ACTH(4-10) fragment rather than a copy of the full hormone. Truncation plus the added tail removes the steroidogenic portion of ACTH. That distinction explains why it is studied for nervous-system effects instead of adrenal stimulation.
It is registered as a prescription product in Russia and has been used there clinically, chiefly by the intranasal route. No approval exists in the United States or the European Union. Availability elsewhere typically falls under research-chemical distribution, with varying purity documentation.
Each freeze-thaw cycle can degrade a fraction of the peptide and promote aggregation. Dividing a stock solution into single-use volumes removes repeated cycling as a variable. It also limits the time a thawed solution spends at room temperature.
